In vitro evaluation of a novel Estradiol analogue with nanomolar antimitotic and carbonic anhydrase inhibitory activity. In the present study we describe the in vitro biological activity of 2-ethyl-3-O-sulphamoyl-estra-1,3,5(10),15-tetraen-17-ol (ESE-15-ol), a novel antimitotic compound with anticarbonic anhydrase activity. This study indicates that ESE-15-ol is a promising antimitotic anticancer drug that warrants further investigation.

Antimitotiese middels is een van die mees gebruikte chemoterapeutiese antikankermiddels wat in die kliniek gebruik word vir die behandeling van kanker. Die doeltreffendheid van die middels teen kanker kan verbeter word deur hul biobeskikbaarheid te verhoog. Die sintese van antimitotiese middels wat omkeerbaar bind aan koolsuuranhidrase II (KAII) in rooibloedselle is een manier om die biobeskikbaarheid en doeltreffendheid te verbeter. Omkeerbare binding aan KAII word gevolg deur ’n stadige vrystelling in die bloedsomloopstelsel. In die huidige studie beskryf ons die in vitro-biologiese aktiwiteit van 2-etiel-3-O-sulphamoïel-estra-1,3,5(10),15-tetraen-17-ol (ESE-15-ol), ’n nuut ontwerpte antimitotiese middel met nanomolare koolsuuranhidrase inhiberende aktiwiteit. ESE-15-ol is in staat om die kanker-verwante koolsuuranhidrase IX (KAIX- aktiwiteit teen ’n laer konsentrasie (89 nM) as die KAII-isoform (167 nM) te inhibeer. Rekenaar-analises met behulp van Autodock Vina het aangedui dat die dehidrasie van die D-ring ’n rol speel in die selektiwiteit van ESE-15-ol na KAIX. ESE-15-ol en was in staat om selgroei te verminder tot 50% na 48 h teen 50 nM – 75 nM in MCF-7, MDA-MB-231 en MCF-12A selle. Die verbinding is die minste kragtig teen die nie-tumorigene MCF-12A selle. In vitro-meganistiese studies het getoon dat die nuut gesintetiseerde verbinding mitochondriale membraan depolarisasie geïnduseer het en die fosforileringstatus van Bcl-2 verander het. Geen-uitdrukking van gene wat verband hou met seldood is ook geïnduseer. Hierdie studie dui aan dat die ESE-15-ol ’n belowende antimitotiese kankermiddel is wat verdere ondersoek regverdig.
